目的:建立高效液相色谱串联质谱法(High-Performance Liquid Chromatography Tandem Mass Spectrometry,HPLC-MS/MS)检测精神病患者血清中的多种抗精神病类药物(Antipsychotic drugs,AD)的浓度.方法:以氯氮平(LDP)、去甲氯氮平(QJLDP)、利培酮(LPT)、帕潘立酮(PPLT)、齐拉西酮(QLXT)稳定同位素为内标,样品处理后进行HPLC-MS/MS上清液分析.结果:各药物携带污染率为-0.17%~2.04%;在各自质量浓度水平表现出现良好的内线性,相关系数均>0.9900;测得批内、批间准确度和精密度,回收率及基质效应均符合性能验证要求.结论:HPLC-MS/MS体系在前处理操作上更加简便,且检测速度更快,具有更高的灵敏度和准确性.这种方法不仅提高了临床检测的效率,同时也为精神病患者的个体化治疗提供了有力的支持.
High performance liquid chromatography tandem mass spectrometry for detecting the concentration of various antipsychotic drugs and metabolites in the serum of psychiatric patients
To establish a high-performance liquid chromatography tandem mass spectrometry(HPLC-MS/MS)method for detecting the concentration of various antipsychotic drugs(AD)in the serum of psychiatric patients.Method:Stable isotopes of chlorpromazine(LDP),norclozapine(QJLDP),risperidone(LPT),papazolidone(PPLT),and ziprasidone(QLXT)were used as internal standards.After sample treatment,HPLC-MS/MS supernatant analysis was performed.ResμLt:The contamination rate of each drug was-0.17%~2.04%;Good internal linearity was observed at their respective mass concentration levels,with correlation coefficients>0.9900;The accuracy and precision,recovery rate,and matrix effect measured within and between batches meet the performance validation requirements.Conclusion:The HPLC-MS/MS system is more convenient in pre-treatment operations,with faster detection speed,higher sensitivity and accuracy.This method not only improves the efficiency of clinical testing,but also provides strong support for personalized treatment of psychiatric patients.
