Application of High Performance Liquid Chromatography in Monitoring the Blood Concentration of Rifampicin in Patients with Pulmonary Tuberculosis
Objective:To analyze the effectiveness of high-performance liquid chromatography in monitoring the blood concentration of rifampicin in patients with pulmonary tuberculosis.Method:100 patients with pulmonary tuberculosis in the hospital were selected from January 2021 to December 2022,all of whom received treatment with rifampicin.High performance liquid chromatography and ultraviolet spectrophotometry were used to monitor blood drug concentration,and the changes in blood drug concentration within 24 hours after use were observed.Based on the monitoring results of blood drug concentration 5 days after use,the patients were divided into a high-level group(>9mg/L)and a low-level group(≤9mg/L),and the levels of inflammatory factors and immune function indicators were compared between the two groups.Results:The linear range of blood drug concentration determined by high-performance liquid chromatography was 0.3~30 μ g/mL.Combined with linear regression equation analysis,it was found that there was a good linear relationship between blood drug concentration and peak area in this range of drugs in plasma(r=0.9941);One patient with low blood drug concentration was not detected by high-performance liquid chromatography,with a total detection rate of 99.00%.Eight patients were not detected by ultraviolet spectrophotometry,with a total detection rate of 92.00%.Moreover,the average analysis time of high-performance liquid chromatography was relatively short(P<0.05).At the time of enrollment,the levels of inflammatory factors were similar between the two groups.Four months after medication,the high blood drug concentration group had lower levels of IL-6,IL-8,CRP,and TNF-α(P<0.05).Conclusion:High performance liquid chromatography has a high effective rate and short time required for monitoring the blood concentration of rifampicin.In addition,the blood concentration of rifampicin in tuberculosis patients can affect the anti-inflammatory effect.
