天然产物五味子乙素对口腔癌细胞增殖、凋亡及侵袭的影响

摘要
目的 研究天然产物五味子乙素(Schisandrin B,Sch B)对口腔癌细胞增殖、凋亡和侵袭的影响.方法 用不同浓度的五味子乙素分别处理口腔癌细胞株SCC15 和CAL27 细胞 24 h和 48 h,采用CCK-8 法和AnnexinV-FITC/PI 双标记流式细胞术,检测五味子乙素对口腔癌细胞增殖及凋亡的影响;采用Transwell实验分别检测五味子乙素对细胞侵袭的影响,采用qRT-PCR和Western Blot检测五味子乙素对Prx1 mRNA与蛋白表达的影响.结果 CCK-8 和流式细胞术检测发现五味子乙素明显抑制SCC15 和CAL27 细胞增殖并促进细胞凋亡,呈时间和剂量依赖性(P<0.05).Transwell实验结果显示,五味子乙素对侵袭的抑制作用呈时间和剂量依赖性.五味子乙素对Prx1 mRNA 的表达无显著影响,但对其蛋白表达有显著的抑制作用(P<0.05).结论 五味子乙素可能通过调控Prx1蛋白的表达抑制口腔癌细胞的增殖、侵袭,促进细胞凋亡.
Abstract
Objective To investigate the effect of schisandrin B(Sch B)on the proliferation,apoptosis and invasion on oral cancer cells.Methods The cell viability was detected by CCK8 assay.Apoptosis was measured by Annexin V-fluorescein isothiocyanate/propidium iodide staining.Cell invasion was tested using transwell matrigel invasion assay.The mRNA and protein expression of peroxiredoxin1(Prx1)was examined by qRT-PCR and Western blot.Results Sch B significantly inhibited the proliferation of SCC15 and CAL27 cells and promoted cell apoptosis in a time-dependent and dose-dependent manner(P<0.05).The inhibitory effect of Schisandrin B on invasion was time-dependent and dose-dependent.Schisandrin B had no significant effect on the expression of Prx1 mRNA,but had a significant inhibitory effect on its protein expression(P<0.05).Conclusions Sch B may inhibit the proliferation and invasion of oral cancer cells and promote cell apoptosis by regulating the expression of Prx1 protein.
