摘要
目的 探讨苦参碱对口腔鳞癌细胞系CAL27 生物学行为的影响.方法 体外培养CAL27 细胞至对数生长期,采用不同浓度苦参碱处理细胞24 h、48 h、72 h,四甲基偶氮唑盐法检测其对细胞增殖的影响并计算半数抑制浓度,划痕实验检测苦参碱对细胞迁移能力的影响,Transwell实验检测苦参碱对细胞侵袭能力的影响,克隆形成实验检测苦参碱对细胞克隆形成能力的影响.结果 应用苦参碱可抑制CAL27 细胞的增殖、迁移、侵袭和克隆形成能力.结论 苦参碱可以抑制人口腔鳞癌细胞增殖、迁移、侵袭、克隆形成能力,有望成为治疗口腔鳞状细胞癌的新型辅助药物.
Abstract
Objective To investigate the effect of Matrine on biological behaviors of CAL27 oral squamous cell carcinoma cell line.Methods In this study,CAL27 cells were cultured in vitro and treated with different concentrations of Matrine for 24 h,48 h and 72 h.Cell proliferation was assessed using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay and the half-maximal inhibitory concentration(IC50)was determined.Additionally,the impact of Matrine on cell migration capacity was evaluated through scratch assays,while its effect on cell invasion capability was detected using Transwell assays.Furthermore,the influence of Matrine on the colony-forming ability of the cells was analyzed through clonogenic formation assays.Results Matrine inhibited the proliferation,migration,invasion,and clonogenic formation ability of CAL27 cells in a concentration-dependent manner.Conclusions Matrine demonstrates the potential to inhibit the biological behaviors of proliferation,migration,invasion and clonogenic formation in human oral squamous cell carcinoma cells.It holds promise as a novel therapeutic agent for the treatment of oral squamous cell carcinoma.
