摘要
目的 研究雷帕霉素对人腮腺间充质干细胞(parotid mesenchymal stem cells,PMSCs)在放疗后的防护作用.方法 雷帕霉素处理人PMSCs后接受放疗,观察其增殖、凋亡、成骨分化能力及细胞外基质中Ⅰ型、Ⅲ型胶原的变化.结果 放疗会造成人PMSCs的增殖率下降、凋亡率升高、成骨分化能力下降.PMSCs加入雷帕霉素后接受放疗,增殖率下降、凋亡率升高、成骨分化能力下降等现象均得到改善.放疗可以使细胞外基质中Ⅰ型胶原增高,Ⅲ型胶原没有变化.提前加入雷帕霉素无法影响细胞外基质中的Ⅰ型胶原、Ⅲ型胶原.结论 放疗导致人PMSCs的干性降低、细胞外基质中I型胶原堆积.放疗前使用雷帕霉素可以改善PMSCs在放疗后干性降低的现象.
Abstract
Objective To investigate the protection of rapamycin on human parotid mesenchymal stem cells(PMSCs)under irradiation-induced injury.Methods PMSCs were cultured in vitro and exposed to one single dose(8 Gy)irradiation.The effect of rapamycin on cell proliferation,apoptosis and osteogenic differentiation ability and changes of collagen I and collagen Ⅲ in extracellular matrix(ECM)were evaluated.Results Irradiation resulted in a decreased proliferation and osteogenic differentiation ability and increased apoptosis rate of PMSCs.Rapamycin protected the proliferation and osteogenic differentiation ability,decreased cell apoptosis after irradiation.Irradiation increased collagen Ⅰ in the ECM,while collagen Ⅲ remained unchanged.Rapamycin did not affect collagen Ⅰ and Ⅲ in the ECM.Conclusions Irradiation reduced the stemness of human PMSCs and promoted the accumulation of collagen Ⅰ in the ECM.Rapamycin improved the decreased stemness of PMSCs after irradiation.
