摘要

目的 评估新型生物陶瓷类材料c-Root SP对人根尖牙乳头干细胞(hSCAPs)的细胞增殖及骨诱导能力.方法 制备不同浓度c-Root SP浸提液,采用CCK-8 法检测其对hSCAPs的细胞增殖情况,使用碱性磷酸酶(ALP)活性测定法测定ALP酶水平和茜素红染色法观察钙化结节形成情况,数据采用单因素方差分析,与iRootSP对比,评价c-Root SP促进细胞增殖及骨诱导能力.结果 c-Root SP对hSCAPs的细胞增殖具有浓度依赖性,1/10 浓度浸提液增殖水平最好,c-Root SP1/5、1/10、1/1000 浓度浸提液对hSCAPs的细胞增殖的影响显著高于同浓度iRoot SP浸提液(P<0.05).成骨方面,矿化诱导 7 天,iRoot SP ALP活性显著高于c-Root SP(P<0.05),矿化诱导 14 天 c-Root SP矿化结节显著多于iRoot SP.结论 c-Root SP具有对hSCAPs的良好细胞增殖及骨诱导能力,与iRoot SP相似.

Abstract

Objective To evaluate the ability of c-Root SP,a new bioceramic materials,on cell proliferation and bone induction in human stem cells from the apical papilla(hSCAPs).Methods Different concentrations of c-Root SP extracts were prepared.The cell proliferation of hSCAPs was detected by CCK-8 method,ALP enzyme level was examined by alkaline phosphatase(ALP)activity method and the formation of calcified nodules was observed by alizarin red staining method.Single factor analysis of variance was used.The cells proliferation and bone induction ability of c-Root SP were evaluated and compared with iRoot SP.Results c-Root SP had a concentration dependent effect on the cell proliferation of hSCAPs,with a 1/10 concentration extract having the best proliferation level.The effects of c-Root SP 1/5,1/10,and 1/1000 concentration extracts on hSCAPs cell proliferation were significantly higher than those of iRoot SP extracts at the same concentration(P<0.05).In terms of osteogenesis,after 7 days of mineralization induction,the activity of iRoot SP ALP was significantly higher than that of c-Root SP(P<0.05),and the mineralization nodules in c-Root SP were significantly more than those in iRoot SP after 14 days of mineralization induction.Conclusions C-Root SP has good cell proliferation and bone induction ability to hSCAPs,similar as iRoot SP.
