Objective To investigate the expression and role of the mechanosensitive ion channel Piezo1 in the compression-side alveolar bone during orthodontic tooth movement(OTM)in rats, and to explore the possible signaling pathways. Methods The OTM model was established, and the expression levels of the mechanosensitive ion channel Piezo1,osteoclasts number, osteoblast markers(RANKL,OPG),osteogenic markers(Runx2,Osterix)and the Wnt non-classical signaling pathway(Wnt5a,CAMKⅡ)were examined in the compression-side periodontal tissues at different time points. Results During OTM process, the osteoclasts number and RANKL expression increased;Piezo1,OPG,Runx2,Osterix,Wnt5a and CAMKⅡ levels decreased significantly after the initial increase. Conclusions Piezo1 may be involved in the remodeling of the alveolar bone on the compression side during OTM process and may function through the Wnt non-classical signaling pathway.
