摘要

为了探讨ASPH6在肺癌发生和发展中的生物学作用,构建了ASPH6真核表达载体,分别转染了2株肺腺癌细胞系LTEP-a-2和NCI-H1299,经G418筛选后获得了稳定的克隆细胞株.体外实验结果显示,表达ASPH6的肺癌细胞与对照空载细胞相比,细胞的迁移和浸润能力明显下降,但细胞的生长无明显改变.动物体内实验性转移结果显示,高表达ASPH6可明显抑制肺癌细胞的转移能力.进一步通过蛋白质组学方法,筛选并鉴定出14个ASPH表达调控蛋白.因此推测,ASPH6很可能是一个在肺癌发生和发展中起负调控作用的基因.

Abstract

To elucidate ASPH6 function and corerelation between ASPH6 and lung cancer, a full length ASPH6 expression vector was constructed and stably transfected into lung adenocarcinoma cell lines LTEP-a-2 and NCI-H1299. After selection with G418, clones stably over-expressing ASPH6 were obtained. In migration assay and scratch wounding, it was found that lung cancer cells over-expressing ASPH6 demonstrated reduced motility and invasive capacity, but no significant changes in cell grow rate were found in comparison with vector control. In vivo tumorigenicity assay in SCID mice showed that ASPH6 over-expression inhibited cancer cell metastasis. Two dimensional electrophoresis and mass spectrum analysis revealed 15 proteins differentially expressed in LTEP-a-2/ASPH6 and LTEP-a-2/Neo cells. Therefore, ASPH6 gene may negatively regulate lung cancer progression.
