Quantitative determination of tetrahydropalmatine in Xiaohuahuangjin (Corydalis racemosa) by HPLC
Objective To establish a method for quantitative determination of tetrahydropalmatine in Xiaohuahuangjin (Corydalis racemosa) by HPLC. Methods The HPLC procedure was performed on the chromatographic column of Gemini C18 (250 mm × 4. 6 mm, 5 μm) at 25℃, the mobile phase was methanol-0. 1% phosphoric acid water solution (75:25). The flow velocity was 1.0 mL/min and the detection wavelength was 280 nm. Results The average content of tetrahydropalmatine in Xiaohuahuangjin was 0. 160% . Tetrahydropalmatine showed a good linear relationship at a range from 0. 193 μg to 2. 128 μg. The average recovery was 98. 55% (RSD = 1. 67%, n =6). Conclusion The method was simple and reliable with a good repeatability, which can be used for the quantitative determination of tetrahydropalmatine in Xiaohuahuangjin.
