Effect of Mongolian medicine total flavones of Dracocephalum moldavica L.on the expression of vascular endothelial growth factor receptor 2 in the ischemic semipenumbra of rats after cerebral ischemia-reperfusion
Objective:To observe the effect of Mongolian medicine total flavones of Dracocephalum moldavica L.(TFDM)on the expression of vascular endothelial growth factor 2 (VEGFR2)in ische-mic penumbra of rats after cerebral ischemia-reperfusion,and to explore its effect on angiogenesis. Methods:The middle cerebral artery occlusion (MCAO)reperfusion model of SD rats was prepared by suture method.TFDM50 mg/kg was administered by intragastric administration.The samples were taken at 1,3,7 and 14 days after operation.Immunohistochemical staining was used to observe the distribution of VEGFR2 positive labeled cells and neovascularization in the ischemic penumbra.West-ern blot was used to detect the expression of VEGFR2 in the ischemic penumbra.Results:Immunohis-tochemical staining showed that the number of positive cells in the TFDM administration group on the 3rd to 14th day after MCAO reperfusion was significantly higher than that in the model group (P<0. 05 ),and the expression increased near the microvessels,arterioles and venules.Western blot also showed that the expression of VEGFR2 was up-regulated in TFDM group.Conclusion:TFDM may upregulate the expression of VEGFR2 protein,promote the generation of microvessels in the ischemic penumbra,and improve microcirculation function.
