Synthesis Optimization of Thiolated Chitosan based on Central Composite Design-Response Surface Method
In order to obtain the product with high biological properties,the synthesis conditions of thiolated chitosan was optimized based on central composite design-response surface method.The ratio of N-acetyl-L-cysteine to chitosan and pH value of reaction solution were selected as the main factors,and the content of free thiol content of the product was used as the evaluation index to pre-dict and verify the model.The results showed the binomial fitting equation has good predictability and the free thiol content of the opti-mized polymer is(505.4±9.8)μmol/g,which is close to the predicted value.Also,the central composite design-response surface method can optimize the synthesis conditions of thiolated chitosan,and the obtained polymer has high content of free sulfhydryl group and mucosal adhesion.
