摘要

目的 对蚱蝉的虫生真菌进行分离鉴定,初步探索其虫生真菌的种类多样性及真菌的抗菌活性.方法 采用组织切块法分离纯化蚱蝉虫生真菌,形态特征学和ITS序列鉴定所得真菌种属,并用MEGA7 软件以邻接法构建系统发育树.以氯霉素为阳性对照药物,滤纸片法筛选具有抗菌活性的真菌.结果 从蚱蝉中分离得 15 株真菌,比对分析确定了 11 株真菌属于 7 属,分为曲霉属、链格孢属、枝孢子虫属、镰刀菌属、青霉属、苜蓿属、亚罗酵母属.编号为ZL8、ZL11、ZL14 对金黄色葡萄球菌、大肠肝菌、变形杆菌、白色念珠菌有显著的抑菌活性,鉴定为Cladosporium halotolerans、Penicillium sp.、Stagonospora sp..结论 研究初步探索了蚱蝉虫生真菌的种类,为其虫生真菌资源和次级代谢产物方面的研究奠定基础.

Abstract

Objective To isolate and identify entomogenous fungi from Cryptotympanaatrata,and explore preliminarily their species diversity and antibacterial activity.Methods Tissue sectioning method was used to isolate and purify the entomogenous fungi of the Cryptotympanaatrata,morphological characteristics and ITS sequence identification were used to obtain the fungal species and genera,and MEGA7 software was used to construct a phylogenetic tree using adjacency method.Chloramphenicol as a positive control drug,filter paper method was used to screen fungi with antibacterial activity.Results Fifteen fungi were isola-ted from the Cryptotympanaatrata,and eleven fungi belonged to seven genera,including Aspergillus sp.,Alternaria sp.,Clados-porium sp.,Fusarium sp.,Penicillium sp.,Stagonospora sp.and Yarrowia sp..ZL8,ZL11 and ZL14 had significant antibac-terial activity against Staphylococcus aureus,Escherichia coli,Proteus and Candida albicans,and were identified as Cladospori-um haloolerans,Penicillium sp.and Stagonospora sp.Conclusion The study preliminarily explored the types of entomophytic fungi in the grasshopper,laying a foundation for the research on their entomophytic fungal resources and secondary metabolites.
