Effect of Wnt4 recombinant protein on osteoblasts under inflammatory state
Objective To establish an inflammatory environment in vitro,compare the effects of Wnt4 recombinant protein on osteoblast differentiation and mineralization ability under inflammatory state,and preliminarily explore the role of Wnt4 recombi-nant protein in regulating osteoblast differentiation under inflammatory state.Methods Osteoblasts were extracted from the crani-al parietal bone of mice by enzyme digestion method. The effects of LPS and Wnt4 recombinant protein on the proliferation and differentiation of mouse osteoblasts were detected by CCK8 method. The concentration gradient was designed and the optimal con-centration was determined according to the detection results. After osteogenic induction culture in vitro,they were divided into four groups according to whether LPS and Wnt4 recombinant protein were added,including the control group,the LPS group,the Wnt4 group,and the LPS+Wnt4 group. Alkaline phosphatase activity was compared after 7 days of culture. After 21 days of culture,alizarin red staining was used to compare cell mineralization ability.Results After7 days of subgroup culture,alkaline phosphatase activity in the LPS+Wnt4 group was significantly higher than that in the LPS group,with P<0. 05. The results of alizarin red staining on the 21st day showed that the mineralization capacity of the LPS+Wnt4 group was higher than that of the LPS group,with P<0.05.Conclusion Wnt4 recombinant protein (0.05 μg/mL)plays a positive role in regulating the differ-entiation of osteoblasts under inflammatory conditions.
