Expression and clinical significance of SIRT6 in gastric cancer
Objective To investigate the expression and clinical significance of Sirtuin 6(SIRT6)in gastric cancer tissues.Methods Bioinformatics was used to analyze SIRT6 mRNA expression in gastric cancer and its relationship with survival,tumor mutational burden(TMB)and TIDE score.Ninety-five cases of gastric cancer and adjacent tissue samples were collected,the expression level of SIRT6 was detected by immunohistochemistry,and the relationship between SIRT6 and clinicopathologic fea-tures was analyzed.SIRT6 expression in 10 cases of fresh gastric cancer and adjacent tissues was detected by Western blotting.Results Bioinformatics analysis showed that SIRT6 mRNA expression in gastric cancer was higher than that in normal gastric tis-sues(P<0.05).Kaplan-Meier survival curve showed that the survival time of the group with high SIRT6 expression was shor-ter.Mutation correlation analysis and TIDE score showed that SIRT6 high expression group had higher TMB and TIDE score(P<0.05),and SIRT6 mRNA expression level was significantly positively correlated with TMB and TIDE score(P<0.05).Immunohistochemical results showed that the expression rate of SIRT6 in gastric cancer tissues was 85.2%(81/95).The high expression of SIRT6 in gastric cancer was correlated with high Ki-67 proliferation index,and the difference was statistically signif-icant(P<0.05),while there were no significant differences in SIRT6 expression among different age,gender,tumor diameter and P53 groups(P>0.05).Western blotting showed that the expression level of SIRT6 in gastric cancer tissues was higher than that in adjacent tissues(9/10),and the difference was statistically significant(P<0.05).Conclusion SIRT6 is highly ex-pressed in gastric cancer tissues,which may be involved in the occurrence and development of gastric cancer by affecting the ge-nomic stability and the proliferation ability of gastric cancer cells.
