Effect of total saponins of Panax notoginseng on proliferation and apoptosis of KGIa cells in acute myeloid leukemia
Objective To observe the effects of total saponins of Panax notoginseng on proliferation and apoptosis of acute myeloid leukemia cells KGIa.Methods CCK-8 assay was used to detect the effect of different concentrations of Panax notogin-seng saponins on the survival rate of KGIa cells after 24 hours,48 hours and 72 hours of treatment.The morphology and apopto-sis of KGIa cells treated with total saponins of Panax notoginseng for 72 hours were observed under inverted microscope and fluo-rescence microscope.Flow cytometry was used to detect apoptosis and cell cycle changes.Results The total saponins of Panax notoginseng could significantly inhibit cell proliferation in a concentration dependent manner.The apoptosis of KGIa cells in the notoginseng total saponins group was significantly increased,and notoginseng total saponins blocked the cell cycle in G0-G1 phase.Conclusion Total saponins of Panax notoginseng can inhibit the proliferation of acute myeloid leukemia cells KGIa,which may be related to cell cycle retardation and induction of apoptosis.
