芦竹(Arundo donax)生长周期短,纤维素及粗蛋白含量高,是一种新型的经济能源饲草,具有广阔的应用前景。TCP是一类植物特有的转录因子,在调控植物生长发育过程中具有重要作用,但目前该基因家族在芦竹中的分布以及生物学功能未见报道。本研究以芦竹为对象,利用生物信息学方法对AdTCP基因家族的基本理化性质、染色体定位、基因间的共线性关系、基因结构、蛋白的保守基序、多序列比对以及与其他物种的亲缘进化关系进行了系统分析,并运用实时荧光定量PCR测定了盐胁迫下7个AdTCP基因的相对表达量。结果表明,芦竹含有37个AdTCP基因,分布在不同染色体上,其蛋白理化性质各不相同,且均为亲水性、不稳定蛋白,多数定位在细胞核中。AdTCP家族基因结构中均只含有外显子,无内含子,共线性分析鉴定出22个片段复制基因对;其家族蛋白包含15个不同的保守基序,序列相似性为30。52%。通过与拟南芥(Arabidopsis thaliana)、水稻(Oryza sativa)TCP基因进行亲缘关系比较,结果发现82个TCPs可以分为TCP-P和TCP-C两大亚家族,其中TCP-P仅包含PCF分支,而TCP-C包含CYC与CIN两个分支。随着盐胁迫浓度的升高,7个AdTCP基因的表达量呈现先上升后下降的趋势,说明AdTCP参与盐胁迫响应过程。本研究为解析芦竹基因功能及创制芦竹新种质奠定了基础。
Gene identification and expression analysis of the TCP gene family of Arundo donax under salt stress
Arundo donax has a short growth cycle and high cellulose and crude protein contents.A.donax is a new economic energy forage with broad application prospects.TCP is a plant-specific transcription factor that plays an important role in regulating plant growth and development.However,the distribution and biological functions of this gene family in A.donax have not yet been reported.In this study,the basic physical and chemical properties,chromosome localization,collinearity,gene structure,conserved motifs of proteins,multiple sequence alignment,and phylogenetic relationships with other species of the AdTCP gene family were systematically analyzed using bioinformatics.The relative expression levels of the seven AdTCP genes under salt stress were determined by real-time fluorescence quantitative PCR.Our results showed that the 37 AdTCP genes were distributed on different chromosomes.The physical and chemical properties of different proteins differ.All proteins were hydrophilic and unstable,and most of them were in the nucleus.The gene structure of this family contains only exons and no introns,and 22 fragment-replicating gene pairs were identified by collinear analysis.The protein family contains 15 conserved motifs with a sequence similarity of 30.52%.Compared with TCP genes of Arabidopsis thaliana and Oryza sativa,82 AdTCPs were divided into two subfamilies,TCP-P and TCP-C,in which TCP-P contained only the PCF branch and TCP-C contained the CYC and CIN branches.With an increase in the salt stress concentration,the expression levels of seven AdTCP genes first increased and then decreased,indicating that AdTCP was involved in the salt stress response.This study could lay the foundation for the analysis of gene function and the creation of new germplasms of Arundo donax.
