Optimization of Extraction Process of Polysaccharides from Sika Deer Antler by Response Surface Methodology
Taking Sika Deer Polysaccharide as the research object,the extraction process was optimized for the first time by the combination of ultrasonic extraction,hot water leaching and aqueous alcohol precipitation. On the basis of the one-factor test,a four-factor,three-level response surface test was carried out using the principle of BBD experimental design. The polysaccharide content was determined by phenol sulfuric acid method,and the purity was detected by UV and FT-IR spectral analysis. The results showed that under the optimal extraction process conditions:ultrasonic liquid to material ratio of 34:1,ultrasonic time of 43 min,extraction temperature of 54 ℃,and extraction time of 2 h,the polysaccharide extraction rate reached 2.02%,and the polysaccharide content of 70.05%,which was free of nucleic acids and proteins,etc.,and had a typical polysaccharide structure,which optimised process for extracting Sika Deer Antler Polysaccharide was shown to be practical and feasible,and the results were better than those of the previous process.
