建立HPLC-PDA联合Box-Behnken响应面法,优化蜂胶片的提取工艺.以提取时间、料液比、甲醇浓度为自变量,以白杨素和高良姜素含量的总评归一值为因变量,色谱柱为Hypersil ODS2 C18 柱(4.6 mm×150 mm,5 μm),流动相为甲醇-0.1%磷酸(58∶42),流速为 1.0 mL/min;柱温为25 ℃,检测波长为268 nm;采用Box-Behnken响应面法优选蜂胶片的提取工艺,并进行验证.最佳提取工艺为30倍量83%甲醇提取,提取10 min.Box-Behnken响应面法优选的提取工艺稳定、可靠,可为蜂胶片的提取和质量控制提供参考.
HPLC-PDA Combined with Box-Behnken Response Surface Method to Optimize the Extraction Process of Propolis Tablets
This study establishes HPLC-PDA combined with Box-Behnken response surface method to optimize the extraction process of propolis tablets.Extraction time,solid-liquid ratio and methanol concentration were independent variables,and the total normalized val-ues of Chrysin and galangin contents were dependent variables.The chromatography was performed on Hypersil ODS2 C18 column(4.6 mm× 150 mm,5 μm),and the mobile phase consisted of methanol-0.1%phosphoric acid(58∶42)and the flow rate is 1.0 mL/min;The column temperature was 25 ℃ and the detection wavelength was 268 nm.The Box-Behnken response surface method was used to optimize the extraction process of propolis tablets and verify it.The optimal extraction process was 30 times 83%methanol extraction and the extraction time was 10 min.The optimal extraction process of Box-Behnken response surface method is stable and reliable,which can provide reference for extraction and quality control of propolis tablets.
