栀子豉汤颗粒对失眠大鼠海马组织中单胺类神经递质和炎性因子的影响

研究栀子豉汤颗粒对失眠症的作用并探索相关机制.通过建立大鼠失眠模型,以地西泮为阳性对照,研究栀子豉汤颗粒对大鼠神经递质(5-HT、DA和NE)与炎性因子(IL-1β、IL-6和TNF-α)含量的影响.将SPF级SD雄性大鼠36只,随机分成6组,分别为对照组、模型组、地西泮组、栀子豉汤颗粒低剂量组、栀子豉汤颗粒中剂量组和栀子豉汤高剂量组,每组6只.造模完成后,分别给予相应浓度药物,对照组和模型组给予同体积的生理盐水,观察各组大鼠的行为学变化及DA、5-HT、NE、IL-1β、IL-6和TNF-α水平.旷场实验结果显示,与对照组相比,模型组大鼠的移动距离显著增加(P<0.05),站立次数及粪便颗粒数增加不明显(P>0.05);与模型组比较,地西泮组和栀子豉汤颗粒高剂量组移动距离和站立次数极显著减少(P<0.01);各组大鼠的粪便颗粒数无统计学差异(P>0.05).与对照组相比,模型组大鼠海马组织中的NE、DA、IL-6、IL-1β和TNF-α含量均极显著升高(P<0.01),5-HT含量极显著降低(P<0.01);与模型组对比,地西泮组和栀子豉汤颗粒高剂量组均能极显著降低大鼠海马组织中NE、DA和TNF-α含量(P<0.01),显著升高5-HT含量(P<0.05).结果表明,栀子豉汤颗粒可明显改善模型大鼠的失眠病症,其对大鼠脑内5-HT、NE、DA、IL-6、IL-1β和TNF-α的调节作用可能是其治疗失眠的作用机制.
Study on Regulatory Mechanism of Zhizichi Decoction on Monoamine Neurotransmitters and Inflammatory Factors in Insomnia Model Rats
This study aims to investigate the effect of Zhizichi Decoction granule on insomnia and explore the relevant mechanisms.By establishing the insomnia model of rats and taking Diazepam as the positive control,the effects of Zhizichi Decoction granule on the content of neurotransmitters(5-HT,DA,NE)and inflammatory factors(IL-1 β、IL-6,TNF-α)were studied 36 SPF SD male rats were randomly divided in-to 6 groups:normal control group,model group,Diazepam group,low,medium and high dose groups of Zhizichi Decoction granule,6 rats in each group.After the completion of modeling,the rats were given cor-responding concentrations of drugs,and the normal group and model control group were given the same volume of physiological saline.The behavioral changes and the levels of DA,5-HT,NE,IL-1 β,IL-6,TNF-α in each group were observed.The results of the open-field experiment showed that the moving distance of the rats in the model group increased significantly(P<0.05)compared with the control group,and the number of standing and fecal pellets increased insignificantly(P>0.05).Compared with the model group,the distance traveled and the number of standing were significantly reduced in the Diazepam group and the high-dose group of Zhizichi Decoction granules(P<0.01),and the number of fecal pellets in each group was not statistically different(P>0.05);compared with the control group,the contents of NE,DA,IL-6,IL-1 β and TNF-α in the hippocampal tissue of rats in the model group significantly in-creased(P<0.01),and the 5-HT content was significantly reduced(P<0.01).Compared with the model group,both the Diazepam group and the high-dose group of Zhizichi Decoction granules significant-ly reduced the content of NE,DA and TNF-α(P<0.01)and increased the content of 5-HT(P<0.05)in the hippocampal tissues of rats.It can be concluded that Zhizichi Decoction granules can significantly improve insomnia in model rats,and its modulating effects on 5-HT,NE,DA,IL-6,IL-1 β and TNF-α in rat brain may be its mechanism of action in treating insomnia.
