Study on Regulatory Mechanism of Zhizichi Decoction on Monoamine Neurotransmitters and Inflammatory Factors in Insomnia Model Rats
This study aims to investigate the effect of Zhizichi Decoction granule on insomnia and explore the relevant mechanisms.By establishing the insomnia model of rats and taking Diazepam as the positive control,the effects of Zhizichi Decoction granule on the content of neurotransmitters(5-HT,DA,NE)and inflammatory factors(IL-1 β、IL-6,TNF-α)were studied 36 SPF SD male rats were randomly divided in-to 6 groups:normal control group,model group,Diazepam group,low,medium and high dose groups of Zhizichi Decoction granule,6 rats in each group.After the completion of modeling,the rats were given cor-responding concentrations of drugs,and the normal group and model control group were given the same volume of physiological saline.The behavioral changes and the levels of DA,5-HT,NE,IL-1 β,IL-6,TNF-α in each group were observed.The results of the open-field experiment showed that the moving distance of the rats in the model group increased significantly(P<0.05)compared with the control group,and the number of standing and fecal pellets increased insignificantly(P>0.05).Compared with the model group,the distance traveled and the number of standing were significantly reduced in the Diazepam group and the high-dose group of Zhizichi Decoction granules(P<0.01),and the number of fecal pellets in each group was not statistically different(P>0.05);compared with the control group,the contents of NE,DA,IL-6,IL-1 β and TNF-α in the hippocampal tissue of rats in the model group significantly in-creased(P<0.01),and the 5-HT content was significantly reduced(P<0.01).Compared with the model group,both the Diazepam group and the high-dose group of Zhizichi Decoction granules significant-ly reduced the content of NE,DA and TNF-α(P<0.01)and increased the content of 5-HT(P<0.05)in the hippocampal tissues of rats.It can be concluded that Zhizichi Decoction granules can significantly improve insomnia in model rats,and its modulating effects on 5-HT,NE,DA,IL-6,IL-1 β and TNF-α in rat brain may be its mechanism of action in treating insomnia.
