Antibacterial Effect and Mechanism of Flavonoid Compound Cudraflavone B on MRSA
To investigate the antibacterial effect and mechanism of flavonoid codpound cudraflavone B on methicillin-resistant staphylococcus aureus(MRSA1001),the minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of cudraflavone B were determined by using the micro broth dilution method,and its time bactericidal curve against MRSA1001 was plotted.Its safety was evalu-ated through hemolytic activity and cytotoxicity tests.Finally,the morphological changes of MRSA1001 af-ter treatment were observed by scanning electron microscopy.The results showed that cudraflavone B ex-hibited a significant inhibitory effect on Gram-positive bacteria,weak bactericidal effect,and showed a certain concentration dependent bactericidal activity against MRSA 1001.No significant cytotoxicity was observed on HEK293 cells,and low hemolytic activity was also observed on rabbit red blood cells,which indicated high safety.Under scanning electron microscopy,bacterial cells treated with cudraflavone B showed collapse accompanied by liquid exudation,indicating that its mechanism of action might be a-chieved by disrupting the bacterial cell membrane,and thereby achieving antibacterial effects.In summa-ry,cudraflavone B can continue to be explored for its value as an antibacterial drug.
