目的 观察一脂肪细胞特异性表达的新基因小脂肪细胞因子1(SMAF1)在小鼠脂肪前体细胞3T3-L1中的异位表达对脂肪细胞增殖和凋亡的影响.方法 构建小鼠SMAF1真核表达载体并稳定转染3T3-L1脂肪前体细胞以获得永久异位表达SMAF1基因的细胞株,使用Western blot证实SMAF1基因的表达.体外培养SMAF13T3-L1脂肪细胞,以MTT法检测SMAF13T3-L1脂肪细胞的增殖;用流式细胞仪对SMAF13T3-L1脂肪细胞进行凋亡分析.结果 与对照相比,SMAF13T3-L1脂肪细胞增殖明显降低,差异有统计学意义(P<0.05);而SMAF13T3-L1脂肪细胞凋亡分析结果与对照无显著性差异. 结论 SMAF1基因在3T3-L1中的异位性表达,对3T3-L1前体脂肪细胞生长增殖有显著的抑制作用,而对细胞凋亡无影响,提示SMAF1在脂肪细胞增殖分化中发挥一定的作用.
Effect of ectopic expression of SMAF1 on proliferation and apoptosis of preadipocyte 3T3-L1
Objective To evaluate the effect of a novel adipocyte-specific expressed gene small adipocyte factor 1 (SMAF1) on proliferation and apoptosis of preadipcyte 3T3-L1. Methods A permanent SMAF1 expression construct was established for stable transfection of 3T3-L1 and formation of SMAF1 3T3-L1 cell line. After invitro cul-turing of cells, both MTT method and flow cytometry were used to detect and analyze the proliferation level and apoptosis progress respectively. Results Compared with control groups, SMAF1 3T3-L1 cell line showed remarkable reduction in proliferation with statistical significance (P<0. 05), while no notable difference in apoptosis between two cell lines. Conclusion The ectopic expression of SMAF1 in 3T3-L1 preadipocyte might suppress proliferation without interfering apoptosis process.
