SF3B1/FOXM1/JUNB轴调控SOX21表达对宫颈癌细胞生物学行为的影响研究

目的 分析剪接因子3B亚基1/叉头框转录因子M1/转录因子活化蛋白激酶B(SF3B1/FOXM1/JUNB)轴调控转录因子21抗体(SOX21)表达对宫颈癌细胞生物学行为的影响。方法 选取2022年3月至2023年12月新疆医科大学附属肿瘤医院收治的50例宫颈癌患者的癌旁组织及癌组织作为研究对象,利用实时荧光定量PCR检测SF3B1、FOXM1、JUNB、SOX21表达;通过Transwell、细胞计数试剂8(CCK8)检测宫颈癌细胞生物学行为(增殖、迁移、侵袭);利用蛋白质印迹法测定SF3B1、FOXM1、JUNB、SOX21蛋白表达。结果 与癌旁组织相比,宫颈癌组织JUNB表达低,FOXM1、SOX21、SF3B1表达高,差异有统计学意义(P<0。05);与si-NC组相比,si-SF3B1/FOXM1/JUNB组0 h OD450值高,侵袭细胞数、迁移细胞数、(24、48 h)OD450值、SF3B1、FOXM1、JUNB低,差异有统计学意义(P<0。05);与OE-NC组相比,OE-SOX21组迁移细胞数、(0、24、48 h)OD450值、SOX21、侵袭细胞数高,差异有统计学意义(P<0。05);与si-SF3B1/FOXM1/JUNB+OE-NC组相比,si-SF3B1/FOXM1/JUNB+OE-SOX21 组 SOX21、SF3B1、FOXM1、JUNB、(0、24、48 h)OD450 值、侵袭细胞数、迁移细胞数高,差异有统计学意义(P<0。05)。结论 SF3B1/FOXM1/JUNB轴通过激活SOX21表达可促进宫颈癌细胞侵袭、增殖、迁移。
Effect of SF3B1/FOXM1/JUNB Axis Regulating SOX21 Expression on Biological Behavior of Cervical Cancer Cells
Objective To analyze the effect of splicing factor 3B subunit 1/forkhead box M1/transcription factor activated protein kinase B(SF3B1/FOXM1/JUNB)axis regulated expression of transcription factor 21 antibody(SOX21)on the biological behavior of cervical cancer cells.Methods The para-cancerous tissues and cancerous tissues of 50 cervical cancer patients admitted to the Affiliated Cancer Hospital of Xinjiang Medical University from March 2022 to December 2023 were selected as the study objects.The expression levels of SF3B1,FOXM1,JUNB and SOX21 were detected by real-time fluorescence quantitative PCR.The biological behaviors of cervical cancer cells(proliferation,migration and invasion)were detected by Transwell and cell counting kit-8(CCK8).The protein expressions of SF3B1,FOXM1,JUNB and SOX21 were determined by Western blot.Results Compared with the adjacent tissues,the cervical cancer tissues showed a lower expression level of JUNB and higher expression levels of FOXM1,SOX21,and SF3B1,and the differences were statistically significant(P<0.05).Compared with the si-NC group,the si-SF3B1/FOXM1/JUNB group exhibited a higher optical density(OD)450 value at 0 h,but lower invasive cell count,migratory cell count,(24,48 h)OD450 values,SF3B1,FOXM1,and JUNB,and the differences were statistically significant(P<0.05).Compared with the OE-NC group,the OE-SOX21 group showed higher migratory cell count,(0,24,48 h)OD450 values,SOX21,and invasive cell count,and the differences were statistically significant(P<0.05).Compared with the si-SF3B1/FOXM1/JUNB+OE-NC group,the si-SF3B1/FOXM1/JUNB+OE-SOX21 group exhibited higher levels of SOX21,SF3B1,FOXM1,JUNB,(0,24,48 h)OD450 values,invasive cell count,and migratory cell count,and the differences were statistically significant(P<0.05).Conclusion SF3B1/FOXM1/JUNB axis can promote the invasion,proliferation and migration of cervical cancer cells by activating SOX21 expression.
