Effect of Wushen Decoction on Myocardial Injury in Mice with Septic Cardiomyopathy
Objective To investigate the effect of Wushen Decoction on the myocardial injury in mice with septic cardiomyopathy.Methods Mice were randomly separated into a control group,a model group,and a model+Wushen Decoction group,with 20 mice in each group.Except for the control group,mice in all other groups were injected with lipopolysaccharide to induce septic cardiomyopathy.The mice in each group were given intragastric administration of normal saline or Wushen Decoction accordingly,once a day for 5 d till modeling.And after modeling,the gavage went on for 7 d.The serum levels of tumor necrosis factor-α(TNF-α),interleukin(IL)-18,IL-1β,reactive oxygen species(ROS),malondialdehyde(MDA),glutathione peroxidase(GSH-Px),creatine kinase-isoenzyme(CK-MB),and cardiac troponin(cTnI)were detected.Hematoxylin-eosin(HE)staining was used to observe the pathological changes in myocardial tissue of the three groups.The number of gasdermin D-N terminal(GSDMD-N)positive cells in myocardial tissue was detected by immunofluorescence staining.Western blot was utilized to detect NOD-like receptor family pyrin domain-containing 3(NLRP3),cysteine aspartic protease-1(Caspase-1),GSDMD-N,IL-18,and IL-1β proteins in myocardial tissue.Results Compared with the control group,the model group and model+Wushen Decoction group showed disorderly arranged myocardial cells,a large amount of inflammatory cell infiltration,increased serum levels of TNF-α,IL-18,IL-1β,ROS,MDA,cTnI and CK-MB,elevated number of GSDMD-N positive cells in myocardial tissue,increased levels of NLRP3,Caspase-1,GSDMD-N,IL-18,and IL-1β proteins,and decreased serum level of GSH-Px(P<0.05).Compared with the model group,the corresponding indicators in the model+Wushen Decoction group showed opposite trends to those mentioned above(P<0.05).Conclusion Wushen Decoction can inhibit inflammatory response,oxidative stress,pyroptosis of cardiomyocytes,and NLRP3/Caspase-1/GSDMD pathway activity in mice with septic cardiomyopathy.
