Effects of Resveratrol on Inflammation and Th1/Th2 Imbalance in Rats with Allergic Rhinitis Through TLR4/NF-κB Pathway
Objective To investigate the effects of resveratrol on the inflammatory response,T-helper 1(Th1)and T-helper 2(Th2)balance,and the regulation of Toll-like receptor 4(TLR4)/nuclear factor-κB(NF-κB)pathway in rats with allergic rhinitis.Methods A total of 50 Sprague Dawley(SD)rats were divided into 5 groups:normal group,model group,resveratrol group,activator group,and activator+resveratrol group according to random number table method,with 10 rats in each group.Except for the normal group,the other 4 groups of rats were given intraperitoneal injection and nasal ovoalbumin drops to replicate the allergic rhinitis model.The rats in the resveratrol group were given gavage of 100 mg/kg resveratrol,the rats in the activator group were injected with TLR4 activator lipopolysaccharide(LPS)3 mg/kg in the tail vein,the rats in the activator+resveratrol group were injected with LPS 3 mg/kg in the tail vein and gavage of 100 mg/kg resveratrol,while the rats in the normal group and the model group were given the same amount of normal saline.The interventions were performed once a day for 7 consecutive days.The behavioral symptom score of each group was evaluated.The expression levels of immunoglobulin E(IgE)and interleukin 4(IL-4)were detected by enzyme-linked immunosorbent assay(ELISA).The percentage of Th1 and Th2 cells was detected by flow cytometry.The mRNA expression levels of TLR4 and NF-κB were detected by real-time fluorescent quantitative reverse transcription polymerase chain reaction(qRT-PCR).And the protein expression levels of TLR4 and phosphorylated NF-κB(p-NF-κB)were detected by western blot.Results Compared with the model group,the resveratrol group showed a decrease in the behavioral symptom score,IgE and IL-4 levels,Th2 cell percentage,TLR4 and NF-κB mRNA expression levels,and TLR4 and p-NF-κB protein expression levels,but an increase in Th1 cell percentage(P<0.05).Compared with the activator group,the activator+resveratrol group showed a decrease in the behavioral symptom score,IgE and IL-4 levels,Th2 cell percentage,TLR4 and NF-κB mRNA expression levels,and TLR4 and p-NF-κB protein expression levels,but an increase in Th1 cell percentage(P<0.05).Conclusion Resveratrol can alleviate behavioral symptoms,reduce inflammatory response and regulate Th1/Th2 cell balance in rats with allergic rhinitis,possibly by inhibiting TLR4/NF-κB pathway.
