摘要

目的 探讨白藜芦醇对变应性鼻炎大鼠炎症反应和辅助性T细胞1(Th1)/辅助性T细胞2(Th2)平衡的影响,以及对Toll样受体4(TLR4)/核因子-κB(NF-κB)通路的调节作用.方法 将50只SD大鼠按照随机数字表法分为正常组、模型组、白藜芦醇组、激活剂组和激活剂+白藜芦醇组5组,每组10只.除正常组外,其余4组大鼠通过腹腔注射和滴鼻卵白蛋白复制变应性鼻炎模型,白藜芦醇组大鼠灌胃白藜芦醇100 mg/kg,激活剂组大鼠尾静脉注射脂多糖(TLR4激活剂)3 mg/kg,激活剂+白藜芦醇组大鼠尾静脉注射脂多糖3 mg/kg,同时灌胃白藜芦醇100 mg/kg,正常组和模型组大鼠同时给予等量生理盐水,5组干预均为1次/d,连续7 d.评估各组大鼠行为学症状评分,酶联免疫吸附测定(ELISA)法检测免疫球蛋白E(IgE)、白细胞介素-4(IL-4)等表达水平,流式细胞仪检测Th1和Th2细胞百分比,实时荧光定量逆转录聚合酶链反应(qRT-PCR)检测TLR4、NF-κB mRNA表达量,蛋白印迹法检测TLR4、磷酸化核因子-κB(p-NF-κB)蛋白表达量.结果 与模型组比较,白藜芦醇组大鼠行为学症状评分、IgE和IL-4水平、Th2细胞百分比、TLR4和NF-κB mRNA表达量、TLR4和p-NF-κB蛋白表达量均降低,Th1细胞百分比升高(P<0.05);与激活剂组比较,激活剂+白藜芦醇组大鼠行为学症状评分、IgE和IL-4水平、Th2细胞百分比、TLR4和NF-κB mRNA表达量、TLR4和p-NF-κB蛋白表达量均降低,Th1细胞百分比升高(P<0.05).结论 白藜芦醇可减轻变应性鼻炎大鼠行为学症状,降低炎症反应,调节Th1/Th2细胞平衡,其作用机制可能是通过抑制TLR4/NF-κB通路发挥作用.

Abstract

Objective To investigate the effects of resveratrol on the inflammatory response,T-helper 1(Th1)and T-helper 2(Th2)balance,and the regulation of Toll-like receptor 4(TLR4)/nuclear factor-κB(NF-κB)pathway in rats with allergic rhinitis.Methods A total of 50 Sprague Dawley(SD)rats were divided into 5 groups:normal group,model group,resveratrol group,activator group,and activator+resveratrol group according to random number table method,with 10 rats in each group.Except for the normal group,the other 4 groups of rats were given intraperitoneal injection and nasal ovoalbumin drops to replicate the allergic rhinitis model.The rats in the resveratrol group were given gavage of 100 mg/kg resveratrol,the rats in the activator group were injected with TLR4 activator lipopolysaccharide(LPS)3 mg/kg in the tail vein,the rats in the activator+resveratrol group were injected with LPS 3 mg/kg in the tail vein and gavage of 100 mg/kg resveratrol,while the rats in the normal group and the model group were given the same amount of normal saline.The interventions were performed once a day for 7 consecutive days.The behavioral symptom score of each group was evaluated.The expression levels of immunoglobulin E(IgE)and interleukin 4(IL-4)were detected by enzyme-linked immunosorbent assay(ELISA).The percentage of Th1 and Th2 cells was detected by flow cytometry.The mRNA expression levels of TLR4 and NF-κB were detected by real-time fluorescent quantitative reverse transcription polymerase chain reaction(qRT-PCR).And the protein expression levels of TLR4 and phosphorylated NF-κB(p-NF-κB)were detected by western blot.Results Compared with the model group,the resveratrol group showed a decrease in the behavioral symptom score,IgE and IL-4 levels,Th2 cell percentage,TLR4 and NF-κB mRNA expression levels,and TLR4 and p-NF-κB protein expression levels,but an increase in Th1 cell percentage(P<0.05).Compared with the activator group,the activator+resveratrol group showed a decrease in the behavioral symptom score,IgE and IL-4 levels,Th2 cell percentage,TLR4 and NF-κB mRNA expression levels,and TLR4 and p-NF-κB protein expression levels,but an increase in Th1 cell percentage(P<0.05).Conclusion Resveratrol can alleviate behavioral symptoms,reduce inflammatory response and regulate Th1/Th2 cell balance in rats with allergic rhinitis,possibly by inhibiting TLR4/NF-κB pathway.
