以鳡鱼肌肉为原料,采用正己烷提取其中的鱼油,通过三因素三水平的Box-Behnken响应面设计法,构建了鱼油得率与提取参数的数学模型并进行预测,获得最优提取参数.并分析了该条件下提取的粗鱼油的理化指标.研究结果表明,在试验范围内,提取次数是主要影响因素,时间和液料比次之;最优提取参数是:提取次数3次,时间30 min,液料比(V∶W)为2∶1(mL/g),平均鱼油得率达(33.64±0.07)%(n=3).此条件下提取的鱼油呈淡黄色、浑浊状态,具有鱼腥味,水分含量为0.34%,杂质0.38%,其中,酸价1.15 mg KOH/g、过氧化值1.84 mmol/kg、碘价135 g I2/100 g,达到了粗鱼油标准SC/T 3502-2000的一级要求.
Optimization of oil extraction from Elopichthys Bambusa muscle by response surface methodology
The oil was extracted from Elopichthys Bam busa muscles by n-hexane through three factors and three levels of Box-Behnken response surface design,the mathematical model between fish yield and extraction parameters was provided and the physical and chemical properties of this fish oil were analyzed.Results showed that the extraction times was the most important factor to fish oil yield,followed by the time and the ratio of solvent to material.The optimum extraction parameters were extraction 3 times,30 min and the ratio of solvent to material (V ∶ W) 2 ∶ 1 (mL/g).The average fish oil yield arrived to (33.64 ± 0.07)% (n =3) when extracted under optimized conditions.The fish oil obtained was light yellow,turbid with fishy smell,and contained 0.34% moisture content,0.38% impurity.Its acid value was 1.15 mg KOH/g,peroxide value was 1.84 mmol/kg,iodine value was 135 g I2/100 g,which reached first grade standard of crude fish oil according to SC/T3502-2000.
