太空诱变对紫花苜蓿耐盐性及离体再生的影响

为了揭示太空诱变对苜蓿耐盐能力的影响,以搭载种子、未搭载苜蓿种子及耐盐品种中苜1号为材料,研究了盐胁迫下苜蓿种子发芽及其再生能力的变化.结果表明,太空诱变后,苜蓿耐盐性与未搭载和中苜1号相比显著增强,250 mmol/L NaCl胁迫下种子发芽率比未搭载增加25.7%,发芽时间缩短,子叶畸变率低.同时,搭载后的种子组培再生能力显著增强,表现为愈伤诱导、胚状体诱导、不定芽形成及根分化的数量和质量显著高于未搭载对照及中苜1号.通过盐胁迫条件下筛选,最终获得3株耐盐变异材料.
Effect of space induction on salt tolerance and plant regeneration of alfalfa
The impact of space induction on salt tolerance of alfalfa was studied by measuring germination rate and seedling regeneration ability with Zhongmu No. 1 alfalfa. Results showed that germination rate of induced seeds increased by 25. 7% ,the aberration rate of cotyledon and germination days decreased compared with that of the CK seeds under stress of 250 mmol NaCl. Space induction showed positive impact on the regeneration ability of alfalfa seedlings under salt stress. Compared with the control,the induced seeds showed higher ability in terms of formation and growth of callus, number of buds and roots. Based on the salt tolerance, 3 mutations were screened out.
