Evaluation of cold tolerance of different Artemisia wellbyi germplasm
[Objective]In order to evaluate the cold resistance difference of 10 Artemisia wellbyi germplasm,[Method]germination tests were carried out in low-temperature light incubators at 20℃(control),16℃,12℃and 8℃using 10 A.wellbyi germplasm at different altitudes.The germination potential,germination rate,root length,bud length and other indexes were measured at different temperatures.The stress physiological indexes were mea-sured at 20℃(control)and 4℃,and the low temperature tolerance ability was comprehensively evaluated by com-bining principal component analysis and membership function.[Result]The results showed that with the decrease of temperature,the germination rate,germination potential,germination index,vigor index,radicle length and germ length of 10 A.wellbyi seeds were lower than those of the control,and there were significant differences in the low temperature tolerance of 10 A.wellbyi seeds.Under low temperature stress,the activities of superoxide dismutase(SOD)and catalase(CAT)and the contents of soluble protein(SP)and soluble sugar(SS)were increased,and the contents of proline(Pro)and peroxidase(POD)activity were different among different materials(P<0.05).[Conclusion]The results of comprehensive evaluation of 7 indexes of principal component screening by membership function method showed that the cold tolerance of No.2 A.wellbyi germplasm was strong,while that of No.37 A.wellbyi germplasm was weak.
