Optimization study on strain cultivation period and infectious concentration on soybean cotyledonary node via Agrobacterium-mediated transformation system
In soybean cotyledonary node transformation, Agrobacterium infection was influenced by many factors, The cultivation period of Agrobacterium strain and infectious concentration were studied in the research. The results showed that the highest rate of resistant buds was obtained when strain cultivation concentration was 0.6 at OD_(600), and the proper infectious concentration was 0.5 at OD_(600). Seven cultivars of soybean in north China were also screened for high susceptibility to Agrobacterium and three genotypes were found to be suitable for transformation. This research lays a good foundation for improvement of soybean transformation.
