鸭梨过氧化物酶基因全长克隆与序列分析

采用RT-PCR和RACE方法,从鸭梨(Pyrus bretschneideri Rehd)果实中获得过氧化物酶(Peroxidase)基因POD的cDNA全长,命名为PbPOD,并将该基因在GenBank上登录,登录号为JQ325052.基因序列分析表明,该基因的cDNA全长为1487 bp,包含72 bp的5'非翻译区、404 bp的3'非翻译区和长度为1011 bp且编码336个氨基酸的编码区(CDS).氨基酸序列分析表明,该基因编码的蛋白具有POD家族保守存在的所有功能活性位点,且与甜瓜、拟南芥、烟草等植物POD相似性达92%以上.
Cloning and sequence analysis of peroxidase gene cDNA from Yali pear
A full-length cDNA sequence of peroxidase gene was obstained from Yali pear using RT-PCR and RACE, named PbPOD. GenBank accession JQ325052, Sequence analysis indicated that cDNA was 1 487 bp in full length and contains 5' untranslated region (5'-UTR) of 72 bp, 3' untranslated region (3'-UTR) of 404 bp, and an coding areas (CDS), encoding a 336 predicted amino acids residues which possessed all the conserved active sites. Sequence alignment of amino acids revealed that PbPOD shared more than 92%homoloy with POD from plants of Cucumis melo L., Arabidopsis thaliana and Nicotiana tabacum.
