Effect of cold resistance on winter wheat seedlings with ABA pouring around root
In order to investigate the physiological and biochemical mechanism of cold tolerance of winter wheat regulating by exogenous ABA in cold area, two winter wheat cultivars Dongnongdong-mai1 (cold resistant) and Jimai22(cold susceptible) in 3-leaf stage were poured with ABA (the content were 0, 0.1, 1, 10, 100 μmol·L-1) around roots. Under field conditions, leaves and tillering nodes of two winter wheat varieties were sampled when the day lowest temperature was 0,-10,-25 ℃ to measure the physiological indices and to investigate the reviving rate of seedlings. The results showed that contents of osmoregulation substance and ABA,and the ratio of ABA/GA in leaves and tillering nodes of two winter wheats were decreased significantly with 100 μmol·L-1 ABA. The GA content was not affected notably in tillering nodes of two winter wheat varieties, which was increased in leaves of Jimai22. The MDA content was decreased, the SOD activity was increased and the membrane per-meability rised in all of them. Contents of osmoregulation substance and ABA,and the ratio of ABA/GA in leaves and tillering nodes of the two cultivars were increased significantly with 10 μmol·L-1 ABA. Meanwhile, the contents of GA and MDA were decreased and the SOD activity was increased, but soluble protein content and membrane lipid peroxidation were not affected significantly in leaves and tillering nodes of Jimai22. Soluble sugar content in leaves and tillering nodes, and ABA content in tillering nodes, and the ratio of ABA/GA were decreased as well in Jimai22. The other concentrations of ABA did not affected membrane lipid peroxidation and soluble proteins content obviously. The results suggested 10 μmol·L-1 ABA could enhance the cold resistance of Dongnongdongmai1 but not Jimai22.
