猪传染性胃肠炎病毒感染PK15细胞抗病毒相关因子转录变化分析

为研究猪传染性胃肠炎病毒感染与固有免疫之间关系,研究构建含有巴马猪IFN-α1, IFN-β和NF-κB启动子的报告基因载体,将转染上述报告基因的PK15细胞接种TGEV和灭活TGEV后发现,与灭活TGEV相比,TGEV能够诱导IFN-α1、IFN-β和NF-κB报告基因低水平表达;其次,利用荧光定量RT-PCR方法检测接种病毒0、2、6和10 h后PK15细胞中IFN-α1、IFN-β、IL6、TNF-α、ISG56和IRF7转录量,结果表明,TGEV对IL6转录没有影响,而诱导IFN-α1、IFN-β、TNF-α、ISG56和IRF7低水平转录,分别为对照组1。49、1。5、2。2、2。5和1。8倍。
Analysis of antiviral cytokines transcriptional profiles of PK15 cells ;cultures following infection with transmissible gastroenteritis virus
In order to survey the relationship between transmissible gastroenteritis virus infection and host innate immunity responses. Promoters of IFN-α1, IFN-β and NF-κB were constructed and then transfected into PK15 cel s infected by TGEV and inactive TGEV. We found that TGEV could induce the weak expression of IFN-α1, IFN-βand NF-κB, compared with inactive TGEV. Furthermore, Real-timeq PCR was carried out to detect the transcript levels of cytokines (IFN-α1, IFN-β, IL6, TNF-α, IRF7 and ISG56) at 0, 2,6 and 10 h. The results showed that the transcription levels of IFN-α1, IFN-β, TNF-α, ISG56 and IRF7 was 1.49, 1.5, 2.2, 2.5 and 1.8 times with TGEV infection compared with inactive TGEV-HX, excepted IL6.
