Analysis of antiviral cytokines transcriptional profiles of PK15 cells ;cultures following infection with transmissible gastroenteritis virus
In order to survey the relationship between transmissible gastroenteritis virus infection and host innate immunity responses. Promoters of IFN-α1, IFN-β and NF-κB were constructed and then transfected into PK15 cel s infected by TGEV and inactive TGEV. We found that TGEV could induce the weak expression of IFN-α1, IFN-βand NF-κB, compared with inactive TGEV. Furthermore, Real-timeq PCR was carried out to detect the transcript levels of cytokines (IFN-α1, IFN-β, IL6, TNF-α, IRF7 and ISG56) at 0, 2,6 and 10 h. The results showed that the transcription levels of IFN-α1, IFN-β, TNF-α, ISG56 and IRF7 was 1.49, 1.5, 2.2, 2.5 and 1.8 times with TGEV infection compared with inactive TGEV-HX, excepted IL6.
