Effects of gossypol on oxidative damage and expression of NF-κB,P53 and Caspase-3 in mice testis
In order to explore the effects of different gossypol concentrations on oxidative damage and the expression of NF-κB,P53 and Caspase-3 in mice testicular tissue,48 adult male mice were randomly divided into four groups,and were respectively been exposing to gossypol by oral according to 0,40,80 and 120 mg·kg-1·d-1 for 20 d.The results showed that the mice appeared obvious poisoning symptoms,the body weight in middle-dose group(MG)and high-dose group(HG)decreased as the time of exposure to gossypol lengthened.The testis indexes in MG significantly decreased(P<0.01)than those in control group(CG).The activities of SOD and GSH-Px significantly decreased.The NF-κB mRNA expression levels in testicular tissues and protein expression levels in total protein and nuclear protein significantly decreased in a dose-dependent manner.Conversely,the mRNA and protein expression levels of P53 and Caspase-3 in testicular tissues all increased dose-dependently as exposure to gossypol.The contents of MDA showed the same increase.It was suggested that oxidative damage induced by exposed gossypol was closely related to apoptosis in mice testis.It followed that reproductive toxicity was associated with enhancing the expression of P53 and Caspase-3 and inhibiting the expression of NF-κB in the apoptosis pathway.
