胃铋镁药物通过自噬促进铋清除的机制研究

胃铋镁药物是临床上治疗幽门螺旋杆菌的一种铋剂药物,为探索胃铋镁药物清除细胞内重金属铋的作用机制,应用原子荧光吸收法、蛋白质免疫印迹、实时荧光定量PCR、MDC(尸丹黄酰胺)染色法等实验探究了胃铋镁药物对于细胞自噬功能的影响.结果表明:胃铋镁组在细胞内铋的残留量是铝酸铋组的20%,胃铋镁组细胞中自噬相关蛋白LC3B-Ⅱ、Beclin1、ATG7、ATG5、ATG16L的蛋白表达升高,p62蛋白表达降低,Beclin1和多种ATG基因转录水平显著升高;同时胃铋镁组细胞中自噬小体的数量明显增多.胃铋镁药物可通过提高细胞自噬水平具有对细胞内重金属铋的显著清除作用.
Study on the mechanism of Weibimei promote bismuth clearance through autophagy
The drug of Weibimei is a kind of antimony drug for the clinical treatment of Helicobacter pylori.The purpose of this study aimed to explore the effect of Weibimei on the obviation heavy metal bismuth in cells and its associated mechanism.Atomic fluorescence absorption method,western blotting,real-time quantitative PCR,MDC(cadaver dandanamide)staining and other experiments were used to explore the effect of gastric bismuth magnesium on the autophagy function of cells.The results showed that the residual amount of bismuth in the gastric bismuth magnesium group was 20%of that in the bismuth aluminate group.The protein expression of autophagy-related proteins LC3B-Ⅱ,Beclin1,ATG7,ATG5 and ATG16L in the gastric bismuth magnesium group was increased followed with the increased and the expression of p62 protein decreased,and the transcription levels of Beclin1 and various ATG genes.At the same time,the number of autophagosomes in gastric bismuth magnesium cells increased significantly.So Weibimei Drug achieved the scavenging effect on intracellular heavy metal bismuth by increasing the cellular autophagy.
