Systematic Identification of miR10193 Family Members and Nodulation Relavant Expression in Soybean
miR10193 is a miRNA group related with nematode infection in soybean.The role of miR10193 in soybean nodule is yet to be discovered.To systematically identify soybean miR10193 family members and analyze their expression patterns during symbiotic nitrogen fixation,in this study,we identified soybean 12 miR10193 family members at the genome-wide level and performed bioinformatics analysis,and then analyze the expression patterns in different soybean tissues and early rhizobia infection by qRT-PCR.We identified 12 miR10193 members in soybean genome,which distributed on chromosomes 3,4,7,8,10,13,16,19 and 20.The mature and star sequences of soybean miR10193 family members are highly conserved.Phylogenetic analysis of miR10193 precursor sequences generated two subfamilies,we found gma-miR10193c and gma-miR10193h had tandem duplication.The cis regulatory element analysis revealed that miR10193 may be involved in plant hormones,stress,growth and development.The digital expression analysis shown multiple miR10193 members were highly expressed in nodules,indicating their potential role in the regulating soybean symbiotic nodulation.The qRT-PCR results showed except gma-miR10193d/j/k/l,most miR10193 family members were highly expressed in nodules,and we found all miR10193 family members significantly respond to rhizobium infection.Target prediction showed that BTB/POZ domain protein and 2OG-Fe(Ⅱ)oxidase family protein might be the target of miR10193.This study will provide theoretical basis for studying the function and mechanism of miR10193 family in soybean symbiotic nitrogen fixation.
