大豆GmHSF21基因的生物信息学分析

植物热激转录因子(Heat shock transcription factor,HSFs)在参与高温、干旱及盐碱等逆境胁迫方面发挥重要作用.为探究大豆GmHSF21基因在盐胁迫应答中的作用,文章以大豆品种中黄13前期盐胁迫处理下的转录组差异基因数据为研究对象,对大豆HSF基因家族中的候选基因GmHSF21进行生物信息学分析.结果表明,GmHSF21属大豆HSF基因家族中的A亚族,共编码341个氨基酸,其编码区存在4处多态性位点,具有保守的HSF和HSF DNA-binding基序.系统发育分析表明,AT3G22830与GmHSF21亲缘关系近.顺式作用原件分析表明该基因启动子区具有响应多种生物及非生物胁迫的作用元件.荧光定量PCR结果显示该基因在盐胁迫条件下表达量上调,与转录组结果基本一致.
Bioinformatics Analysis of GmHSF21 Gene in Soybean
Plant heat shock transcription factors(HSFs)played important roles in participating in stress conditions such as high temperature,drought,and salinity.To explore the role of GmHSF21 gene in salt stress response,this article took transcriptome differential gene data of soybean variety Zhonghuang 13 under pre salt stress treatment as the research object,and conducted bioinformatics analysis on candidate gene GmHSF21 in the HSF gene family.The results showed that GmHSF21 belonged to subgroup A of the HSF gene family,it encoded 341 amino acids.The coding region had four polymorphic sites,conserved HSF and HSF DNA-binding motifs.Phylogenetic analysis showed that AT3G22830 was closely related to GmHSF21.The analysis of cis-acting elements showed that the promoter region had acting elements in response to various biological and abiotic stresses.Fluorescence quantitative PCR results showed that the expression of the gene was up-regulated under salt stress,which was basically consistent with the transcriptome results.
