Objective:To investigate the association between osteoporosis and type 2 diabetes based on bioinformatics.Methods:The gene expression omnibus database was used to obtain the datasets associated with os-teoporosis and type 2 diabetes and identify the differentially expressed genes of osteoporosis-type 2 diabetes,and the data were imported into STRING database to identify the key genes.The single cell sequencing dataset was used as the validation set to validate the expression levels of genes at the cellular level,and the gene ontology functional en-richment analysis and the Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis were performed for related results.Frequency statistics and functional classification were performed for key targets and related tradi-tional Chinese medicine(TCM)drugs.Results:The common pathways of osteoporosis and type 2 diabetes includ-ed the ErbB signaling pathway,the MAPK signaling pathway,the neurotrophin signaling pathway,and the nucleotide-binding oligomerization domain-like receptor signaling pathway.Frequency statistics showed that 51 TCM drugs were associated with the key genes of osteoporosis and type 2 diabetes,and according to their function,they were classified into heat-clearing and detoxicating drugs,pain-relieving drugs,drugs for dispelling pathogenic wind,deficiency-tonifying drugs,and antiemetic drugs.Conclusion:Both osteoporosis and type 2 diabetes are com-plex metabolic diseases,involving various gene expression networks and signaling pathways.Although both diseases have a complex gene network background,analysis results show that there are still highly coincident differentially expressed genes and signal pathways between the two diseases.This suggests that there is a close association in mo-lecular mechanism between the two diseases and there may be common regulatory targets;therefore,these targets may become therapeutic targets for drug intervention of the two diseases at the same time.
