Effect and mechanism of action of compound traditional Chinese medicine emulsion gel in treatment of acne
Objective:To investigate the pharmacodynamic action of compound traditional Chinese medicine(TCM)emulsion gel on a rabbit ear acne model and its preliminary mechanism of action.Methods:Oleic acid was used to establish a rabbit ear acne model,and drug intervention was performed after successful modeling.HE stai-ning of histopathological sections was performed to analyze the therapeutic effect of compound emulsion gel;ELISA was used to measure related inflammatory factors,including tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6),and interleukin-8(IL-8);immunohistochemistry was used to measure the ex-pression levels of the key signaling pathways mediating the development of acne,including nuclear factor-kappa B(NF-κB),leukocyte differentiation antigen(CD36)receptor,and NOD-like receptor protein 3(NLRP3)inflam-masome.Results:HE staining and immunohistochemistry showed that there was a significant improvement in rabbit ear acne lesions after treatment with compound emulsion gel,and the compound emulsion gel group had significant reductions in the expression levels of the inflammatory factors TNF-α,IL-1β,IL-6,and IL-8 in serum(P<0.05 or P<0.01).Compound emulsion gel could mediate the key signaling pathway NF-κB in the development of acne and downregulated the expression levels of CD36 receptor and NLRP3 inflammasome(P<0.05 or P<0.01).Con-clusion:The compound TCM emulsion gel exerts a therapeutic effect on acne by inhibiting Propionibacterium acnes,downregulating the expression of TLRs receptor and CD36 receptor on various types of cells associated with acne,blocking the NF-κB pathway,inhibiting the activation and expression of NLRP3 inflammasome,and reducing the expression levels of the inflammatory factors TNF-α,IL-6,IL-8,and IL-1β in serum.
