Effect of Liuwei Dihuang decoction on bone fusion after lumbar fusion surgery in a rabbit model:An experimental study
Objective:To investigate the effect of Liuwei Dihuang decoction on bone fusion after lumbar fusion sur-gery in a New Zealand rabbit model.Methods:A total of 30 New Zealand rabbits were randomly divided into blank group,sham-operation group,control group,positive drug group,and traditional Chinese medicine(TCM)group,with 6 rabbits in each group.The New Zealand rabbits in the TCM group,the positive drug group,and the control group were used to estab-lish a model of lumbar intertransverse process fusion;after 7 days of modeling,the rabbits in the TCM group were given the solution of Liuwei Dihuang decoction at a dose of 10 mL/kg by gavage,those in the positive drug group were given an equal volume of calcium carbonate solution by gavage,and those in the control group and the sham-operation group were given an equal volume of 0.9%sodium chloride injection by gavage,twice a day for 8 consecutive weeks.The rabbits in all groups were sacrificed after the end of administration,and the serum concentrations of alkaline phosphatase(ALP),bone morphoge-netic protein 2(BMP2),and procollagen I N-terminal propeptide(PINP)were measured,while HE staining was performed for the sections of fused bone.Results:After drug intervention,the TCM group and the positive drug group had significantly higher serum levels of ALP,BMP2,and PINP than the control group,the sham-operation group,and the blank group(P<0.05 or P<0.01),and the TCM group had significantly higher levels than the positive drug group(P<0.01).HE staining showed that compared with the positive drug group and the control group,the TCM group had better coupling between the bone graft and the host bone and a higher active degree of osteoblasts,with a slighter inflammatory response.Conclusion:In the New Zealand rabbit model after lumbar fusion surgery,Liuwei Dihuang decoction can effectively improve the levels of the bone formation markers ALP,BMP2,and PINP,promote the coupling between the implanted bone and the host bone,improve the active degree of osteoblasts,and alleviate inflammatory response at the site of bone graft fusion.
