Effect of acupuncture on behavioristics in a rat model of depression
Objective:To investigate the effect of acupuncture on behavioristics in a rat model of depres-sion.Methods:A total of 24 male rats were randomly selected to establish a model of depression by social isolation combined with chronic unpredictable mild stress,and after successful modeling,they were divided into model group,fluoxetine group,and acupuncture group,with 8 rats in each group.Another 8 normal male rats were randomly se-lected as blank control group.The rats in the blank control group and the model group were given 0.9%sodium chloride injection by gavage at 9:00 every morning;in addition,the rats in the acupuncture group were given acu-puncture at Baihui and Guanyuan points,and those in the fluoxetine group were given fluoxetine drug solution 1.8 mg/kg by gavage at 9:00 every morning.On days 1,7,14,21,and 28 of intervention,the Morris water maze test was performed for the rats in each group to record the number of times of self-modification,region visit,follow-ing a partner,and body contact,and the rats were observed in terms of behavioristics and learning and memory abili-ties.Results:After intervention,the model group,the fluoxetine group,and the acupuncture group had a significantly shorter latency than the blank control group(P<0.05),and on day 21 of intervention,the fluoxetine group and the acupuncture group had a significantly shorter latency than the model group(P<0.05 or P<0.01).On days 7 and 14 of intervention,the model group,the fluoxetine group,and the acupuncture group had a significantly lower num-ber of platform crossings in the water maze test than the blank control group(P<0.05),and there was a significant difference between the acupuncture group and the model group on days 1,14,21,and 28 of intervention(P<0.05 or P<0.01).Compared with the blank control group,the model group,the fluoxetine group,and the acupuncture group had significantly lower numbers of times of following a partner,self-modification,partner body contact,and region visit(P<0.05),and there were significant differences in these indicators between the fluoxetine group and the model group on day 14 of intervention(P<0.05 or P<0.01).Conclusion:Acupuncture can regulate and im-prove behavioristics and learning and memory abilities in rats with depression.
