Effect of Panax notoginseng saponins on the proliferation and apoptosis of pulmonary artery smooth muscle cells induced by monocrotaline and the ADAM10/Notch3 signaling pathway
Objective:To investigate the effect of Panax notoginseng saponins(PNS)on the proliferation and apoptosis of rat pulmonary artery smooth muscle cells and the ADAM10/Notch3 signaling pathway under the ac-tion of monocrotaline(MCT).Methods:PASMCs cultured in vitro were randomly divided into normal group(group C),MCT group(group M),and MCT+PNS group(group P),and CCK-8 assay was used to measure cell viability;the EdU method was used to measure cell proliferation;qPCR and Western blot were used to measure the mRNA and protein expression levels of ADAM 10,Notch3,Hes-1,p27,proliferating cell nuclear antigen(PCNA),and caspase-3.Results:Under the action of MCT,there were increases in the viability of PASMCs,EdU positive rate,and the mRNA and protein expression levels of ADAM 10,Notch3,Hes-1,and PCNA(P<0.05,P<0.01),and there were reductions in the mRNA and protein expression levels of p27 and caspase-3(P<0.05,P<0.01),while PNS could reduce the above effects under the action of MCT(P<0.05,P<0.01).Conclusion:PNS can weaken the proliferation of rat PASMCs and promote their apoptosis by inhibiting the ADAM10/Notch3 signaling pathway.
