Mechanism of action of Polygonum cuspidatum-Clematis chinensis drug combination in treatment of gouty arthritis:A study based on network pharmacology and experimental validation
Objective:To investigate the mechanism of action of the Polygonum cuspidatum-Clematis chinensis drug combination in the treatment of gouty arthritis based on network pharmacology and experimental validation.Methods:The TCMSP platform was used to obtain the main active components and action targets of the Polygonum cuspidatum-Clematis chinensis drug combination,GenneCards database was used to predict the targets associated with gouty arthritis,and Venn diagram was plotted to obtain the intersecting targets between the Polygonum cuspidatum-Clematis chinensis drug combination and gouty arthritis.Cytoscape 3.7.1 was used to construct a traditional Chinese medicine-component-disease-target regulatory network,and STRING database was used to construct and analyze the protein-protein interaction network.R language and Bioconductor platform were used to perform gene ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis,and animal experiments were con-ducted for validation.Results:The Polygonum cuspidatum-Clematis chinensis drug combination exerted a therapeutic effect on gouty arthritis through 12 active components,26 action targets,and 152 biological pathways.The main targets in-cluded IL-1B,CCL2,and MMP9,and the key pathway was the IL-17 signaling pathway.After intervention with the Polyg-onum cuspidatum-Clematis chinensis drug combination,there was a tendency of reduction in the content of IL-17 in syno-vial fluid(P>0.05).Conclusion:The Polygonum cuspidatum-Clematis chinensis drug combination exerts a therapeutic effect through multiple components,targets,and pathways,possibly by reducing the level of IL-17.
