Medication rule and mechanism of action of traditional Chinese medicine in treatment of intestinal carbuncle:A study based on data mining and network pharmacology
Objective:To investigate the medication rule and potential mechanism of action of traditional Chinese medicine(TCM)in the treatment of intestinal carbuncle based on data mining and network pharmacology. Methods:The medical records of intestinal carbuncle in Chinese Hundred Hundreds of Traditional Chinese Medicine Clinicians Series were summarized, and Traditional Chinese Medicine Inheritance Support System was used to analyze drug nature, taste, and meridian entry.TCMSP platform was used to identify the active compo-nents of drugs and their targets, and several databases were used to obtain disease targets. The protein-protein inter-action analysis, the gene ontology(GO)functional enrichment analysis, and the Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed, and molecular docking was used to validate the binding between drugs and targets. Results:A total of 102 prescriptions were included, and most drugs had a cold, warm, bitter or sweet taste and mainly entered the spleen and liver meridians. The GO functional enrichment analysis and the KEGG pathway enrichment analysis showed that these drugs exerted an anti-inflammatory effect on intestinal carbuncle mainly by regulating inflammatory factors and participating in the tumor necrosis factor and interleukin-17 signaling pathways.Molecular docking showed close binding between quercetin/kaempferol and core targets.Conclusion:Blood-activating and stasis-resolving therapy and heat-clearing and detoxicating therapy are the main TCM therapies for intestinal carbuncle, and its potential mechanism of action mainly involved the process of inflammatory response, which provides a scientific basis for TCM treatment of intestinal carbuncle.
