Analysis of composition of intestinal bacterial genera based on 16S rRNA sequencing technology in children with infectious diarrhea
Objective To analyze the composition of intestinal bacterial genera in children as different areas and different ages in Xinjiang,based on 16S rRNA high-throughput sequencing technology.Methods A total of 194 fecal samples were collected from children patients in the department of outpatient/emergency and inpatient in Urumqi Children*s Hospital from April to November 2023.The DNA of pathogen was extracted and sequenced.The cluster and diversity analysis were carried out,and the intestinal flora spectrum was plotted in the children of Xinjiang for analyzing the differences of intestinal bacterial genera of children in different areas and different ages.Results There were 15 886 809 valid sequences and 91 736 operational taxonomic units(OTUs)were obtained by 16S rRNA high-throughput sequencing.The composition of intestinal microbiota in children was in the order of Firmicutes,Actinobacteria,Proteobacteria,Bacteroidetes,Verrucomicrobia,and Fusobacteria at the phylum level,and Bifidobacterium,Escherichia,Bacteroides,Enterococcus,Streptococcus,and Akkermansia at the genus level.There were no statistical differences between the indexes of Shannon(t=0.867),Simpson(Z=0.878),Ace(Z=-0.740)and Chao(Z=-0.566)in northern and southern Xinjiang(P>0.05).There were statistical differences(P<0.05)in the indexes of Shannon(F=16.627),Ace(H=24.300),Chaol(H=28.570),and SimPson(H=18.323)in three age groups.Conclusions There are differences in intestinal bacterial genera among children in different areas and age groups in Xinjiang.The intestinal microbiota show diversity in children.
