Screening of carotenoids mutants in Rhodosporidium toruloides
A mutant strain of oil synthesis , defective strain or carotenoid was constructed by mutagenesis of plasma and guanidine .It is determined that the pathway of lipid synthesis is the dominant branching pathway of acetyl coenzyme A node through the distribution of acetyl coenzyme A flux .Compound mutagenesis was carried out using plasma and nitroso guanidine .XP-1 ,XO-3 ,XY-4 and XR-2 mutants were screened out through ARTP and NTG mutagenesis with 95% and 90% lethal rate .The ability of producing lipid ,composition of fatty acid and content of carotenoid determination results showed that the carotenoid contents of XP-1 ,XO-3 ,XY-4 and other three strains were lower than that in strain NP11 , the metabolic pathway of carotenoids have been destroyed , but the decreasing of carotenoid could not promote the increasing of lipid synthesis .The carotenoid content increased with the decreasing of lipid content in strain XR-2 ,which could reached to 0 .77 μg/g .The results indicated that acetyle-CoA is a weakly rigid node ,and the pathway of lipid synthesis is the dominant branch of the pathway . The reduction of its products can increase the production of carotenoid synthesis .
