The integration efficiency of heterologous gene mediated by transposons in Saccharomyces cerevisiae
To compare and analyze the effects of different transposons on the integration efficiency of exogenous genes in Saccharomyces cerevisiae,G418 expression cassettes were constructed,mediated by transposons Ty1,Ty2,and Ty4,as well as recombinant strains BY-Ty1,BY-Ty2 and BY-Ty4 of BY4741.It was found that the average colony count of BY4741 recombinant strains expressing G418 mediated by Ty2 was 122 and the integration efficiency was 93.33%,both of which were higher than those of Ty1 and Ty4.G418 resistant recombinant strain BY-Ty2 mediated by Ty2 has stronger resistance to G418 than BY-Ty1 and BY-Ty4,with higher gene copy numbers.The results indicated that transposon Ty2 was more suitable for mediating exogenous gene integration in Saccharomyces cerevisiae.
