Study on Improving Effect of Ginsenoside Rg1 on Spermatogenesis in Mice
Objective To study the protective effect of ginsenoside Rg1 on dibutyl phthalate(DBP)-induced spermatogenic injury in mice and to provide a theoretical basis for the development of natural spermatogenic drugs.Methods Thirty C57BL/6 male mice were ran-domly divided into three groups:control group,DBP group(400 mg/kg)and Rg1+DBP group(Rg1 20 mg/kg,DBP 400 mg/kg).After con-tinuous administration for 35 days,the mice were killed,and serum,testis and epididymis were collected.Sperm density,motility and mal-formation rate were detected and calculated,and levels of testosterone(T),follicle-stimulating hormone(FSH)and luteinizing hormone(LH)in serum were detected by enzymelinked immunosorbent assay.The contents of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and malondialdehyde(MDA)in mouse testis were determined.Results Compared with the control group,the sperm motility and density in the DBP group were significantly decreased(P<0.01),and the sperm malformation rate was not significantly changed.T and LH in serum were significantly decreased,and FSH in serum was significantly increased(P<0.01).The levels of GSH-Px and SOD in testicular tissue were significantly decreased,and the content of MDA was increased(P<0.01).Compared with the DBP group,sperm motility and density in the DBP+Rg1 group were significantly increased(P<0.01),but the sperm malformation rate was not significantly changed.T and LH in serum were significantly increased,and FSH in serum was significantly decreased(P<0.01).The levels of GSH-Px and SOD in testicular tissue were significantly increased,while the content of MDA was significantly decreased(P<0.01).Conclusion Ginsenoside Rg1 could improve DBP-induced reproductive function injury,and the mechanism may be related to the antioxidant effect of ginsenoside Rg1.
