Study on Immunogenomic Typing and Characteristics and Clinical Signi fi cance of Pancreatic Cancer
Objective To establish the immune classification of pancreatic cancer through genomics and further explore its immune char-acteristics and clinical significance.Methods The gene expression profile and clinical characteristics data of 178 patients with pancre-atic cancer were downloaded from the Cancer Genome Map Database,and the patients were grouped by single sample gene set enrich-ment analysis(ssGSEA)score;The immune microenvironment and the composition of immune cells in pancreatic cancer immunotyping samples were evaluated by ESTIMATE and CIBERSORT respectively;Using Kaplan-Meier to plot survival curves and conducting log-rank tests for inter-group differences;GSEA identifies GO enrichment pathways for different immune subtypes.Result Patients were di-vided into high,medium,and low immune groups based on matrix score,immune score,and tumor purity,with statistical differences in prognosis among the three groups.The high immune group contains the highest number of immune cells and stromal cells,while the tumor purity is the lowest;The expression of HLA,PD-L1(CD274),and CTLA4 was significantly increased;Plasma cells,activated CD4+memory T cells,CD8+T cells,and γ8 The content of T cells is the highest,while MO macrophages and M2 macrophages are the least;The overall survival rate was significantly lower than that of patients in the low immune group;The anti immune activity was significantly in-creased.Conclusion Immunophenotyping of pancreatic cancer is helpful to judge the prognosis of patients.Patients in the high immune group have higher anti-immune activity and lower survival rate.
