Effective method study of fishing and spreading based on the large resin semi-thin sections of rice coleoptile tissue
Conventionally,semi-thin sections are lifted from a water tank and placed on glass slides by a scooping ring,and then stained and observed after baking the slices.However,the traditional extracting and spreading method usually causes the fold and watermark of these sections for the preparation of large sections of the rice coleoptile tissue due to the large section area and the surface tension of water.In this work,the slices were lifted from the grooving liquid by a scooping pen that replaced the scooping ring.The slices were then placed on the scooping liquid with a concentration of 0.001%or 0.0005%Tween20,which were dyed after being dried.Based on this method,smooth slices without folds,watermarks and pollution-free were obtained with complete cell structures.By comparing the folds in Tween20,Chaps and SDS,we found that the fold in Tween20 was the most flat and non-polluting,while SDS had wrinkles and dye residue,and Chaps could only ensure the local developing flatness.
