Several factors affecting isolated microspore culture in broccoli
Isolated microspores of ten different broccoli hybrids were cultivated; embryoids had been obtained from six hybrids, the inducement ratio up to 60%. Among these hybrids, Luzhou 808 had 13. 6 embryos per bud. The optimum stage for the microspore cultivation was from uninucleate anaphase to dikaryolization phase. It was advantageous for embryo inducement by adding suitable a-mount of sucrose, activated carbon, 6-BA, AgNO3 and 2,4-D into medium 1/2NLN. The heat-shock treatment at temperature 32 ℃ for 24h had a positive effect on the embryogenesis. While 2-5 days before plucking buds, the temperature between 15 ℃ and 25 ℃ was suitable for the microspore growth.
