摘要
取长约0.5 mm,带有1-2个叶原基太子参茎尖,接种在MS+0.5 mg·L-1 NAA+0.5 mg·L-1BA+30 g·L-1蔗糖+6.5 g·L-1琼脂培养基上培养40 d,组培苗出苗率为86.7%;接种在1/2MS+1.5 mg·L-1 IBA+0.5 mg·L-1 IAA生根培养基,组培苗生根率最高,为93.3%;组培苗成活率为85.2%,定植成活率为100%.采用目测法和指示植物鉴定法检测,初步认为茎尖组培苗不带病毒.小区对比试验表明,茎尖脱毒苗单株产量比种根组培苗增产102.7%.
Abstract
Pseudostellaria heteriphilla shoot-tip with 0.5 mm in length and one or two leaf primordiums was cultured on MS +0.5 mg·L-1 N AA +0.5 mg·L-1 BA+30·L-1 sucrose + 6.5 g·L-1 agar medium for 40 days and the plantling emergence was 86.7%. On 1/2MS + 1.5 mg·L-1 IBA +0.5 mg·L-1 IAA medium, the highest rooting of 93.3% was obtained. The survival was 85.2% , and the field planting was 100% . The ocular estimation and indicator plant were used to test and preliminarily prove that shoot-tip cultured plantlings were virus-free. According to the plot varietal yield test, the yield per plant from virus-free shoot-tip cultured plantlings was increased by 102.7% as compared with the plantlets derived from stem segments.
